He played college football at Oregon State and was the starting quarterback of the Beavers from 2011 to 2014. In Pleasanton, California, Mannion attended Foothill High school. Not only did Mannion play football as a quarterback, but he also played as a pitcher in his junior and senior year on the baseball varsity team and he also played in the junior varsity basketball team, but he never made the varsity team.

Mannion was chosen by the St. Louis Rams in the third round of the 2015 NFL Draft. In 2015, he began his career as the third quarterback behind Case Keenum and Nick Foles during his rookie year.

Later in 2016 season, In Week 17, on New Year's Day, Mannion completed 3 out of 6 passes for 19 yards with one interception after the starter, Jared Goff was subbed out of the game, losing 44-6 to the Arizona Cardinals.

In 2017,  head coach Sean McVay rested most of his playoff starters and had the playoff seed locked in, Mannion started the regular-season finale against the San Francisco 49ers.
