Mattison was picked by the Minnesota Vikings in the third round of the 2019 NFL Draft. He signed a four-year deal worth $3.4 million. In his first NFL game, he ran for 49 yards on nine carries. His debut touchdown came in Week 3 against the Oakland Raiders.

By the end of his rookie season, he had 462 yards and one touchdown on 100 carries, along with 10 receptions for 82 yards. He also had four carries for 20 yards in the playoffs.

In 2020, he achieved a career-high of 112 yards on 20 carries in Week 5 against the Seattle Seahawks, adding three receptions for 24 yards. Alexander suffered a sprained right ankle during the Vikings' Week 14 victory in Las Vegas and has been unable to practice, leading to him missing the subsequent game against the Cincinnati Bengals in Week 15. As of December 14, 2023, he was ruled out for the upcoming game in Cincinnati.

However, the Vikings' head coach, Kevin O'Connell, indicated that Mattison's availability for the following game was not completely ruled out. His status for Week 16 will be determined at a later time.
