Sermon enrolled at Sprayberry High School and played for the school's team. He committed to the University of Oklahoma for college football. 

During his freshman season, Trey played in all 14 games, starting three of them. That season, he had  744 yards on 121 carries with five touchdowns. The following season, he played with Kennedy Brooks, rushing for 947 yards on 164 carries and 13 touchdowns. During his junior season, he played nine games. He concluded the season with 54 carries for 385 yards and four touchdowns.

For the senior season, he transferred to Ohio State University as a graduate student. In the 2020 Big Ten Championship Game against Northwestern University, his 331 rushing yards were the most in a single game by an Ohio State Player, which is also the highest in a Football Bowl Subdivision championship game.  In the 2021 National Championship Game against Alabama, he was injured in the first game and did not return. 

In the 2021 NFL Draft, the San Francisco 49ers picked him in the 3rd round with 88th.
