He was then hired by Bob Stoops as an offensive coordinator for the Oklahoma Sooners on January 12, 2015. At Oklahoma, Riley led Sooners in his very first season to the country's 7th ranked team, and to the College Football Playoff. He also received the Broyles Award, being named the best assistant coach of the nation.

Bob Stoops retired as head coach on 7 June 2017 and Riley was named his replacement. Riley, anchored by Heisman winner Baker Mayfield, led his team to the Big 12 conference championship in the 2017 season, won second place in the CFP rankings, and guided Sooners to the Rose Bowl CFP semifinals where Oklahoma lost the 2018 Rose Bowl to Georgia 54–48 in double overtime.
