Cheick Diallo attended Our Savior New American School in Centereach, New York where he played for his high school basketball team. In 2013, Cheick was invited to the NBPA Top 100 camp and was his first MVP from Africa.

After completing high school, Cheick Diallo enrolled at the University of Kansas and played there. Cheick missed his freshman season's first five games with Kansas after the NCAA failed to clear him for eligibility reasons.

On November 25, 2015, after the NCAA finally ruled Cheick had received a limited amount of impermissible benefits and was cleared to play for the Jayhawks in their match against Loyola on 1 December.

In his solitary season in Kansas, Cheick averaged 3.0 points and 2.5 rebounds per game in 7.5 minutes. In April 2016, Diallo declared his final three years of college eligibility for the NBA draft.

Cheick played off the bench in just under 31 minutes and scored 19 points against the Los Angeles Clippers in the 133–105 loss of the Pelicans on 10 December 2016.

During his rookie season, he had several tasks with the NBA Development League's Austin Spurs, Long Island Nets and Greensboro Swarm, in accordance with the flexible assignment rules. Cheick scored 14 points during the season in a loss of 116–97 to the Wizards of Washington on 9 March 2018.
