Holmes was an undrafted player of the 2011 NFL Draft and was signed by the Minnesota Vikings as an undrafted free agent. He did not get to play in official matches during his rookie season.
Andre Holmes was born on June 16, 1988, in Hoffman Estates, Illinois to parents Mary and Kevin Holmes. He was a student of James B. Conant High School where he played for his high school football team as a wide receiver. After graduation, he enrolled in the Hillsdale College, where he played college football. During his senior years at college, he set the single-season school record with 104 receptions and over 1,300 receiving yards.

He became the wide receiver of the Buffalo Bills playing alongside players like Charles Clay, Jordan Mills, and Jeremiah Sirles. He was able to record his first touchdown as a Bill on September 10, 2017, during the season-opening match against the New York Jets.
Holmes is still in the roster of the Buffalo Bills but was placed on injured reserve on December 26, 2017, with a neck injury.
Andre Holmes is a married man and is married to Diana Holmes. Holmes met his wife during his time at Hillsdale College as Diana was also a student there. The two have been together ever since. Diana graduated from college with a degree in Biology.
After her graduation, Diana moved to Texas with Andre while he played for Dallas Cowboys. She worked as an Olive Garden waitress for short time. She later moved to England with Andre and worked as a high school swim coach
